Swiping and holding will make the home button act like a scroll bar, and you can rapidly flip through Recent Apps thumbnails. Swiping to the right on the home button will open Recent Apps and launch the previous app. Swiping up the screen halfway twice will open Recent Apps on the first swipe and the app drawer on the second swipe. Swiping halfway up the screen will open Recent Apps, and swiping all the way up the screen will open the app drawer. ![]() Tapping on the button will, as usual, go Home, while long-pressing will open the Google Assistant. Turn on gesture controls, and the Recent Apps buttons will disappear, back will occasionally disappear, and you'll get an elongated home button. By default, Android P has the normal system UI bar, but if you go to settings ⇒ system ⇒ gestures ⇒ "swipe up from home button," you can enable the new gesture system. Internally, the new gesture buttons and new Recent Apps interface are referred to as "Quick Step." Remember, this is a developer preview, so things are not finished, and sure enough the current state of gesture controls needs a lot of work. The home screen looks great, with a simple one-button layout, but everywhere else you see the ugly, lopsided back button. If everything in Google Material has rounded corners, and the display has rounded corners, shouldn't the recent app thumbnails have rounded corners? Gesture controls are a hot mess Google came clean about its new design language at Google I/O, called the "Google Material Theme," but it doesn't seem like it was applied to this screen. I doubt it could be changed through a third-party launcher (it probably needs system-level permissions), but it is a bit more flexible. Recent Apps living in the Pixel launcher app means the interface could now be updated through the Play Store. Before, the Pixel launcher was only a small stub application that enabled the Google apps' launcher functionality, but now the Pixel launcher is actually gaining functional code. Keep in mind, most of the launcher code isn't in the Pixel launcher but is instead in the Google App. The code for Recent Apps now lives in the Pixel Launcher on DP2. One interesting part of the new Recent Apps is that it is no longer part of the system UI. Long-pressing on the input box in Gmail actually selects the placeholder label text (things like "Subject" and "to" in Gmail), and there is never an option to "paste." It's a developer's preview, and things are broken. I hope the plan is to make it super easy to copy and paste text from one thumbnail to another, but right now pasting into an input box doesn't really work. You can long-press on any text to bring up the usual text selection controls, and you can also long-press on a picture in Google Photos and copy it. Not only can you read the thumbnails, though, you can also interact with them. If you want to quickly reference something, the bigger, high-quality thumbnails are nice. The larger thumbnails bring the added bonus of being able to easily read text in a thumbnail. In P, they don't have any title bars at all and instead have an icon overlaid on the top edge. Thumbnails used to get a title bar with an app icon in Android O. Thumbnails are a lot bigger, and below the horizontal row of thumbnails is a search bar and a row of suggested app icons. You now have to tap on an icon and pick "split screen."Īndroid P brings big changes to Recent Apps, which scrolls horizontally now instead of vertically. Split screen doesn't work via long-pressing on the Recent Apps button or via dragging app thumbnails to the side of the screen.

![]() I’ve had more creative ideas than ever and I think a lot of that comes from spending so much time in solitude. I feel like the one upside of quarantine has been that my creativity has been out in full force. ![]() Introverts need their alone time to be creative. I had thought it would be less than that, I don’t know! It was reassuring to know that a great deal of us (one in every three people!) are actually introverts. What I Learned About Introverts About a third of us are introverts! This book is interesting because it explores all of the positive things about introverts, and how much we as a society lose from undervaluing them. We’ve moved to what Cain calls a “culture of personality” where being charismatic and magnetic can often be valued over intelligence and even character. If you prefer working in solitude, or prefer listening over speaking, you may find your efforts underappreciated… especially at work. The book explores what Cain calls “the extrovert ideal,” where our society values charisma and teamwork. Quiet by Susan Cain has been on the best seller list for more than 5 years now. I love what an academic approach she takes to writing the book – there is a lot of history and science involved! Quiet: The Power of Introverts In A World That Can’t Stop Talking Quiet by Susan Cain Summary And while Cain writes about what’s happened to make introversion fall out of style, she (an introvert) explains why both personality types are so important. The reality is that the world needs both introverts and extroverts to survive. I think for me the biggest thing was that reading this book made me feel so incredibly understood – it also made me realize that some of those things that I have always thought are “bad” about my personality aren’t actually all that bad. If friends were around and I was always reading they always commented on my body language, curious as to what the heck I was reading and why I was so engrossed. I was so drawn in that I found myself nodding and shaking my head for most of the book. Usually with non-fiction, I will slog through, reading slowly… but that was not the case with this book. (It sounded boring TBH.) But once I finally started it, I could not put it down. She had recommended this book to me ages ago (this Fall maybe? Before that?) but I kept putting it off. I’ve been meaning to talk more about this book for ages but it wasn’t until quarantine (and having a lot more time to just… think) that I remembered my idea for a post about this book.Ī few months ago, upon Carly’s recommendation, I picked up Quiet. ![]() In my experience, books like that are few and far between, but my answer to that question would be (hands down!) For the generation for CAR and TCR T-cell therapies, RNP electrotransfer can simply be applied about 1 day prior to standard viral transduction of CAR and TCR transgenes.ĬRISPR can be used to engineer T cells to overcome key pathways of immunosuppression that lead to T cell–intrinsic resistance of cancer to ACTs. Importantly, applying this technique is feasible within the constraints and timelines of clinical-grade cell therapy manufacturing in compliance with Good Manufacturing Practices, and incorporating a CRISPR gene-editing step is not expected to increase overall production time ( 33). Cas9 delivered via transfection is degraded within 48 to 72 hours, which limits off-target editing and concerns about downstream Cas9 immunogenicity ( 32). Electrotransfer of RNP complexes into primary T cells can be accomplished efficiently without sacrificing cell viability, resulting in highly efficient gene knockout that is observable within about 48 hours. These challenges were overcome with the development of methods to transfect primary T cells with Cas9 ribonucleoprotein (RNP) complexes comprising recombinant Cas9 protein bound to a gRNA ( 28). For an in-depth comparison of CRISPR, ZFNs, and TALENs, readers are referred to a review by Khan ( 25). Recent technological advances have substantially reduced the risk of off-target mutagenesis with CRISPR, as discussed later in this review. Specificity is a key safety consideration for clinical applications as off-target mutagenesis poses a risk of oncogenic transformation in genetically engineered cells. Finally, the efficiency and specificity of genome editing via CRISPR are generally superior to those of ZFNs and TALENs ( 25, 26). This enables (i) rapid design and implementation of new CRISPR gene knockout/knock-in strategies, (ii) efficient multiplexing via incorporation of multiple gRNAs, and (iii) application of whole-genome CRISPR libraries for high-throughput screening. In contrast, the CRISPR system can be designed to target virtually any genomic sequence simply by altering the gRNA sequence. ZFNs and TALENs have limited potential for multiplexed genome editing and are not amenable to the generation of large-scale libraries ( 25). As a result, new ZFN or TALEN proteins must be engineered for each new target site, and design restrictions (particularly for ZFNs) limit the number of sites that can be targeted efficiently ( 24). ZFNs and TALENs rely on protein–DNA binding to guide site-specific DNA cleavage. CRISPR: Why and HowĬRISPR offers a number of notable advantages over earlier genome-editing technologies, such as zinc finger nucleases (ZFN) and transcription activator–like effector nucleases (TALEN). ![]() Emerging applications of CRISPR in non-T cell–based ACTs are also described. The purpose of this review is to detail recent applications of CRISPR that have advanced the field of cellular therapy, beginning with a discussion of specific genetic manipulations that hold the potential to enhance T-cell potency, safety, and scalability and ending with a review of CRISPR screening efforts that have unveiled novel targets for cell therapy enhancement. ![]() ![]() CRISPR genome-editing techniques are at the forefront of this revolution, offering a versatile platform for applications ranging from multiplexed gene knockout to site-specific transgene insertion to genome-wide genetic screening. Furthermore, genetic engineering approaches that enable use of allogeneic cell sources hold the potential to substantially improve the scalability of cell therapy manufacturing. This has initiated the next generation of cellular products that harbor creative genetic manipulations to improve T cells' potency and safety profiles and that can mitigate immunosuppressive triggers in the tumor microenvironment. As genome-editing technologies have become increasingly accessible, their use in cellular therapy has expanded greatly. ![]() ![]() For example, merchants like 'amazon' will be able to make the payment for you without a need to be redirected to another service like VISA or Paypal. If a member of the Society were to be killed off in an old issue, there was more pathos because we knew the character's inner thoughts and feelings, whereas today the death of a Serpent Society member means as little to the reader as the death of a nameless thug in a Hollywood action flick. Compare that to a modern Serpent Society appearance, where one or two characters, like Sidewinder or Black Mamba, do all the talking while the other members just stand around looking menacing. With thought bubbles, the reader is able to know the intimate inner thoughts of any character, whether the main hero, a member of the hero's supporting cast, one of the villain's henchmen, or even a random bystander.įor instance, when the Serpent Society would appear in an old Captain America comic, the writer could use thought bubbles to show us what each member of the Society was thinking at any given moment, giving depth to otherwise forgettable characters like Cottonmouth and Fer de Lance. ![]() ![]() Thought bubbles are a unique facet of the comics medium that has unfortunately been sacrificed in an attempt to make comics more cinematic.
